Is it possible to run an alteryx workflow without closing the excel input file?

Hi @KarenaGuo you can however you need this Excel Legacy Driver installed and selected in your Input tool in order to do that.

Another quick option would be to cache the input data tool first (right click and select "Cache and Run Workflow") before opening up the excel file again. This should allow you to keep the file open whilst running the workflow. However if you need to edit the tool or if you edit the excel file, you will have to clear the cache and then cache the tool again.
